Connect to RDS instance via Internet or VPN/Direct
In your RDS instance Route Table:
1. If you would like to access RDS via public internet you will have to modify the route 0.0.0.0/0 to use Internet gateway
2. If you would like to use VPN/Direct Connect for connections to your RDS you will have to modify the route 0.0.0.0/0 to use Transit gateway
